Phase 3 Trial Results for First Oral Proteasome Inhibitor NINLARO (ixazomib) Published in The New England Journal of Medicine

Pivotal TOURMALINE-MM1 Results Demonstrated that the Addition of Oral Ixazomib to Lenalidomide and Dexamethasone Significantly Extended Progression-Free Survival, with Limited Additional Toxicity in Patients with Relapsed/Refractory Multiple Myeloma

(BUSINESS WIRE)-- Takeda Pharmaceutical Company Limited (TSE: 4502) today announced that results from the international, randomized, double-blind, placebo-controlled TOURMALINE-MM1 Phase 3 clinical study, evaluating once-weekly oral NINLARO® (ixazomib) capsules plus lenalidomide and dexamethasone versus placebo plus lenalidomide-dexamethasone in patients with relapsed and/or refractory multiple myeloma, have been published in the prestigious New England Journal of Medicine (NEJM). NINLARO was recently approved by the U.S. Food and Drug Administration (FDA), based on the pivotal TOURMALINE-MM1 data, in combination with lenalidomide and dexamethasone for the treatment of patients with multiple myeloma who have received at least one prior therapy.

TOURMALINE-MM1, a double-blind, placebo-controlled trial including 722 patients, is the first Phase 3 study with an oral proteasome inhibitor. As reported in NEJM, the trial results demonstrated a statistically significant and clinically meaningful (35%) extension of PFS (HR 0.74; p = 0.01; median PFS: 20.6 months vs. 14.7 months in control group; median follow-up 14.7 months). A benefit in PFS was observed with the ixazomib regimen across pre-specified patient subgroups, including patients with poor prognosis, such as elderly patients, those who have received two or three prior therapies, those with advanced stage disease, and those with high-risk cytogenetic abnormalities. Overall response rates were 78% in the ixazomib arm versus 72% in the placebo group and at least very good partial response rates were 48% versus 39%. The median time to response was 1.1 months in the ixazomib arm versus 1.9 months in the placebo group, and median duration of response was 20.5 versus 15.0 months, respectively. In the ixazomib and placebo groups, frequencies of serious adverse events (47% vs. 49%) and on-study deaths (4% vs. 6%) were similar; 74% and 69% of patients experienced grade ≥3 adverse events. Grade 3 and 4 thrombocytopenia was more frequent in the ixazomib (12 and 7%) vs. placebo group (5 and 4%). Rash occurred more frequently in the ixazomib group than in the placebo group (36% vs. 23% of the patients), as did gastrointestinal adverse events, which were predominantly low grade. The incidence of peripheral neuropathy was 27% in the ixazomib group and 22% in the placebo group (grade 3 events occurred in 2% of the patients in each study group, and no grade 4 events were reported). Data from TOURMALINE-MM1 were previously presented at the 57th Annual Meeting of the American Society of Hematology (ASH) in Orlando, Florida.

NINLARO is currently under review by the European Medicines Agency (EMA). Takeda has also submitted applications for approval of ixazomib to additional health authorities around the world.

Important Safety Information

WARNINGS AND PRECAUTIONS

    Thrombocytopenia has been reported with NINLARO. During treatment, monitor platelet counts at least monthly, and consider more frequent monitoring during the first three cycles. Manage thrombocytopenia with dose modifications and platelet transfusions as per standard medical guidelines. Adjust dosing as needed. Platelet nadirs occurred between Days 14-21 of each 28-day cycle and recovered to baseline by the start of the next cycle.
    Gastrointestinal Toxicities, including diarrhea, constipation, nausea and vomiting, were reported with NINLARO and may occasionally require the use of antidiarrheal and antiemetic medications, and supportive care. Diarrhea resulted in the discontinuation of one or more of the three drugs in 1% of patients in the NINLARO regimen and < 1% of patients in the placebo regimen. Adjust dosing for severe symptoms.
    Peripheral Neuropathy (predominantly sensory) was reported with NINLARO. The most commonly reported reaction was peripheral sensory neuropathy (19% and 14% in the NINLARO and placebo regimens, respectively). Peripheral motor neuropathy was not commonly reported in either regimen (< 1%). Peripheral neuropathy resulted in discontinuation of one or more of the three drugs in 1% of patients in both regimens. Monitor patients for symptoms of peripheral neuropathy and adjust dosing as needed.
    Peripheral Edema was reported with NINLARO. Monitor for fluid retention. Investigate for underlying causes when appropriate and provide supportive care as necessary. Adjust dosing of dexamethasone per its prescribing information or NINLARO for Grade 3 or 4 symptoms.
    Cutaneous Reactions: Rash, most commonly maculo-papular and macular rash, was reported with NINLARO. Rash resulted in discontinuation of one or more of the three drugs in < 1% of patients in both regimens. Manage rash with supportive care or with dose modification.
    Hepatotoxicity has been reported with NINLARO. Drug-induced liver injury, hepatocellular injury, hepatic steatosis, hepatitis cholestatic and hepatotoxicity have each been reported in
    < 1% of patients treated with NINLARO. Events of liver impairment have been reported (6% in the NINLARO regimen and 5% in the placebo regimen). Monitor hepatic enzymes regularly during treatment and adjust dosing as needed.
    Embryo-fetal Toxicity: NINLARO can cause fetal harm. Women should be advised of the potential risk to a fetus, to avoid becoming pregnant, and to use contraception during treatment and for an additional 90 days after the final dose of NINLARO.

ADVERSE REACTIONS
The most common adverse reactions (≥ 20%) in the NINLARO regimen and greater than the placebo regimen, respectively, were diarrhea (42%, 36%), constipation (34%, 25%), thrombocytopenia (78%, 54%; pooled from adverse events and laboratory data), peripheral neuropathy (28%, 21%), nausea (26%, 21%), peripheral edema (25%, 18%), vomiting (22%, 11%), and back pain (21%, 16%). Serious adverse reactions reported in ≥ 2% of patients included thrombocytopenia (2%) and diarrhea (2%).

SPECIAL POPULATIONS

    Hepatic Impairment: Reduce the NINLARO starting dose to 3 mg in patients with moderate or severe hepatic impairment.
    Renal Impairment: Reduce the NINLARO starting dose to 3 mg in patients with severe renal impairment or end-stage renal disease requiring dialysis. NINLARO is not dialyzable.
    Lactation: Advise women to discontinue nursing while on NINLARO.

DRUG INTERACTIONS: Avoid concomitant administration of NINLARO with strong CYP3A inducers.

SES will pay USD 20 million to increase its fully diluted ownership of O3b from 49.1% to 50.5%, bringing its aggregate equity investment in O3b to date to USD 323 million (EUR 257 million). On completion, SES will consolidate O3b’s net debt, which is currently USD 1.2 billion. The transaction is expected to generate returns exceeding SES’s hurdle rates for infrastructure investments.

O3b Networks – a unique, game-changing global solution

O3b Networks operates a constellation of 12 High Throughput Satellites (HTS) in a Medium Earth Orbit (MEO) around 8,000 kilometres from the Earth. The company offers customers a ‘fibre in the sky’ solution, with each of the constellation’s beams capable of delivering up to 1.6 Gigabits per second (Gbps) of throughput at a low latency of less than 150 milliseconds, a significant improvement over geostationary connectivity.

Since beginning commercial operations in September 2014, O3b has become the fastest growing satellite network in history in terms of capacity contracted. The company now serves more than 40 Enterprise, Mobility and Government clients across 31 countries. To date, over 50% of customers have already upgraded their initial service commitments, demonstrating the attraction of O3b’s unique and ‘game-changing’ solution. Consequently, O3b has a fully protected contract backlog of USD 350 million. O3b has procured an additional eight satellites to accommodate rapidly-expanding demand, with four satellites expected to be launched during H1 2018, and the remaining four satellites expected to be launched in H2 2019. These procurements will increase the size of the current fleet from 12 to 20 satellites (including three satellites currently flying as in-orbit back-up). At ‘steady-state’ utilisation, which is targeted to be achieved by the end of the third year of a satellite’s commercial service, the full operational constellation is expected to generate annualised revenue of between USD 32 million and USD 36 million per satellite.

O3b is expected to generate revenue of more than USD 100 million in 2016, nearly doubling the revenue recorded in 2015. The company’s operational and financial performance trends are currently consistent with the criteria that SES requires to be satisfied before moving to control, including significantly improving EBITDA. The acquisition is expected to become free cash flow accretive to SES (before financing activities) by 2018 and become net income accretive to SES by 2018/2019.

As part of the IFRS purchase accounting treatment, the transaction will give rise to recognition of a gain of approximately USD 500 million relating to the remeasurement to fair value of the current non-controlling interest in O3b. The final amount will depend on the closing date of the transaction.

SES’s BBB/Baa2 investment grade credit rating is expected to be re-affirmed by Standard & Poor’s and Moody’s. Having agreed to acquire a controlling share of O3b, with the ensuing consolidation of the company’s debt, SES will assess over the coming months the optimisation of the enlarged group’s funding structure. SES is committed to maintaining its progressive dividend policy.

Purchase Agreement

The Board of O3b Networks has agreed to evaluate an Initial Public Offering (IPO) process for the remaining 49.5% of O3b shares following receipt of the requisite regulatory approvals, and subsequent completion of SES’s increase in ownership to 50.5%.

From 29 April 2016, and subject to O3b not having launched an IPO, SES has a call option to acquire all of the remaining 49.5% of shares in O3b for a baseline sum of USD 710 million, adjusted for an interest charge, calculated at market rates, to reflect the timing of the potential exercise of the call option. Any decision which SES may take to exercise the call option will be subject to satisfying the company's stated investment hurdle rates. Should such an exercise be relevant, and depending on the timing, potential financing sources, including capital markets, will be evaluated to determine the most appropriate financing option, in keeping with the company's commitment to maintaining SES’s investment grade credit status (BBB/ Baa2).

A put option, effective from 1 October 2017 and subject to O3b not having launched an IPO, is held by the other shareholders of O3b, under which SES would have the obligation to acquire all of the remaining shares in O3b for a baseline sum of USD 710 million, adjusted for an interest charge, calculated at market rates, to reflect the timing of the potential exercise of the put option.

If you think urinary tract infections won’t affect you, think again. According to researchers, approximately 60 percent of women will experience one UTI in their lifetimes and one in four women will suffer a recurrence within six months1, 2.

Eurovision Partners with SES to Broadcast Rio 2016 Olympics

As part of the agreement, Eurovision will lease several transponders on the NSS-806 satellite between July and August 2016 to enable broadcasters across the globe to provide extensive coverage of the Olympics in Rio and distribution of other major sporting events.

NSS-806, which is located at the prime orbital position of 47.5 degrees West, has excellent coverage over Brazil, and was one of the key satellites that were utilised for the delivery of the 2014 FIFA World Cup to audiences throughout North America, Latin America, Europe and Asia.

Al Harithi Inspects Accident Readiness of “Abu Dhabi Police Emergency”

Brigadier Eng. Hussein Ahmed Al Harithi, Director General of Central Operations at Abu Dhabi Police General Headquarters (GHQ), visited the Emergency and Public Safety Department, to inspect the level of readiness of staff members and the different vehicles and techniques used to deal with accidents, and provide awareness, first aid, rescue and public safety to society in general.

During the inspection visit, Brigadier Al Harithi examined the workflow at the department, the different tasks and duties it undertakes, and the speed of response to accidents. He also reviewed the department’s efforts in providing support to other police authorities in similar cases. Brigadier Al Harithi urged staff members to pursue their dedicated efforts and outstanding achievements in a way that keeps pace with the leading stature attained by the Ministry of Interior and the rapid development achieved by the UAE Police in all areas of police work. He also called upon them to further endeavor in the delivery of services that meet the requirements of customers and enhance their satisfaction.

Brigadier Al Harithi was briefed by Colonel Mohammed Ibrahim Al Ameri, Head of the department, about the readiness of staff members and the different vehicles used in rescue operations, in response to the plans of Abu Dhabi Police, which aim to reduce accidents, educate the public, and provide prevention advice and guidance on the proper ways to handle the various types of accidents.

Brigadier Al Harithi toured the various branches and sections, and met with the work teams at the department. At the Technical rescue and Rapid Intervention section, Brigadier Al Harithi reviewed the rescue truck, which includes 350 pieces of rescue equipment. He also inspected the readiness of the UAE Search and Rescue; the Water Rescue teams, and the Hazardous Materials Accidents Response Unit. He also examined the different vehicles and tools used in rescue operations, in accordance with the best international standards; as well as the maternity and heavy weight ambulances.

At the Innovations Exhibition in the Department, Brigadier Al Harithi examined the outstanding innovations provided by the department’s staff members, including the rescue arm kit; the rescue motorcycle, which is equipped with basic rescue equipment and devices used to pull injured people from wells and deep holes, as well as the rescue cameras that are humidity and shock resistant and used in narrow and hard-to-reach spaces.

The Director General of Central Operations also toured the Operations Room, which includes the Ambulance team’s specialized platform. He also visited the UAE Search and Rescue Team’s scale models exhibition, which included replicas of earthquakes and landslides accidents that are used in the training process to raise the efficiency of team members.

He also attended a presentation about the electronic program that links the police with hospitals, and allows health care givers to assess the condition of the patient before arriving to the emergency room, and ensure a high degree of readiness depending on the case.

Brigadier Al Harithi met with Colonel Mohammed Ibrahim Al Ameri, Head of the Emergency and Public Safety Department and reviewed the department’s achievements, work processes, and international and regional awards it has obtained; particularly the international rescue diving center certification and the environment and occupational health awards.

Colonel Al Ameri indicated that the department constantly provides international training and education opportunities to its staff members, by sending them abroad to pursue further academic attainment in disciplines related to their respective work.

Visa Opens its First Innovation Center in Asia

Visa Readies Network of Innovation Centers to Foster Collaborative Payment Innovation on a Global Scale

(BUSINESS WIRE)-- Visa Inc. (NYSE:V) today announced the opening of its Singapore innovation center, the first in a network of new regional innovation centers that Visa is launching globally. The new 7,000 square foot Singapore facility will serve as a destination for clients, partners and developers, across the region, to work alongside Visa experts and jointly create the next generation of payment and commerce applications.

This builds off the success of Visa’s flagship innovation center, One Market Center, which was opened in July 2014, and is located at the company’s headquarter in San Francisco. Additional innovation centers will be opened in other regions later this year, and in 2017.

Accessible to both local and global clients, the new Singapore innovation center will provide Visa’s partners with access to Visa APIs and software developer kits (SDKs) available through the company’s Visa Developer Platform. A hands-on, real-time work space staffed by Visa technologists and business leaders, the center will operate as a commercial gateway, helping Visa’s partners to build market-relevant solutions that extend their service offerings into Asia.

Olympic Games Travel Forecast

Visa projects that Brazil will host an estimated 400,000-500,000 international visitors during the month of August, based on trends observed in recent summer editions of the Olympics. Visa data points to 2016 inbound travel to Brazil increasing by an estimated 1.2 million travelers for the year, compared to the 2010-2015 average, primarily due to preparations for the Olympics Games.

The largest share of travelers to Brazil are forecast to come from Latin America and the Caribbean region (45 percent of all international travelers), followed by Europe (30 percent) and North America (15 percent).

Based on historical data from past Olympic Games, Brazil can expect to see an increase in per traveler spending. During the London 2012 Olympic Games, spending averaged $1,830 per traveler1, which was twice as much per visit than regular tourists to the United Kingdom in 2012.

ADP Launch Community-Based Initiative for Blind People about the Dangers of Drugs

The Criminal Investigation Department at Abu Dhabi Police recently launched a community-based initiative that is considered the first of its kind for visually impaired people and their families to raise their awareness about the dangers of drugs. The initiative avails of the “Braille” system that is widely used by blind people to read and write, in order to combat this scourge, protect community members from falling prey to drugs and extend a helping hand to addicts.

The Abu Dhabi Police received 100 Braille awareness booklets from the Blind Printing Press Center of the Zayed Higher Organization for Humanitarian Care and Special Needs. These booklets are designed to acquaint visually impaired and blind people with the dangers of drugs. Both parties collaborated to issue these booklets to become a beacon of knowledge for this category of people with special needs. The Criminal Investigation Department, represented by the Anti-Drug Section, provided the awareness and educational content, while the Security Media Department carried out the technical supervision and the cover page based on the media identity. The Blind Printing Press Center printed the booklets in Braille using its own specialized printing machines. The booklets will be distributed to the relevant competent authorities, universities, and institutions wishing to acquire them.

Colonel Sultan Suwayeh Al Darmaki, Head of the Anti-Drug Section at the Criminal Investigation Department (CID), said: “This community-based initiative comes in line with the ‘Hope Plant’ initiative, implemented as per the instructions of Lt. General HH Sheikh Saif Bin Zayed Al Nahyan, Deputy Prime Minister and Minister of Interior to provide support and care to people with disabilities and allow them to enjoy a decent life.”

Colonel Al Darmaki noted that this humanitarian initiative, which was adopted by the Anti-Drug Section at the Criminal Investigation Department, aims to inform community members, especially blind people, about the dangers of drugs and the resulting problems, in addition to the dangers of this scourge that destroys the future of young people and addicts. He also highlighted the importance of this police initiative, which embodies the mission undertaken by both, the Abu Dhabi Police and the Zayed Higher Organization, to provide support and care to people with special needs and integrate them into society.

Moreover, Colonel Al Darmaki noted that the initiative aims at educating and raising awareness of blind people and their families about the dangers of drugs. He said: “Media campaigns, seminars, and public meetings usually focused on sighted people. However, there was a need to raise awareness of blind people and their families about the dangers of drug use and abuse, and draw community members and institutions’ attention to the need to protect people with disabilities against all types of dangers, being an important social category.”

Furthermore, Colonel Al Darmaki stressed the keenness of the Anti-Drug Section to make the most of its potential to provide optimal and superior services to the public, especially people with special needs and their families; as part of the plans aimed at reinforcing support for people with special needs. “This approach is in response to the directives of the higher leadership, which dedicates significant attention to supporting, integrating, and empowering this segment, as it is an integral part of the fabric of the UAE society,” he noted.  He also indicated that such initiatives contribute to reinforcing social ties and strategic partnerships between social institutions and blind people.

They also commended the different learning methods, aids and practices provided for the blind, using the «Braille» system, as well as the advanced technical devices and special equipment to support the integration of this category into the society. The delegation members toured the Blind Printing Press Center and were briefed on the printing services it provides and the special programs it undertakes under the supervision of qualified sighted and visually impaired staff members.

Nae’ma Abdul Rahman Al Mansouri handed over Colonel Al Darmaki 100 copies of the drug awareness brochures designed primarily for use by blind and visually-impaired people. Colonel Al Darmaki pointed out to the importance of the initiative in upgrading the level of services provided to the public, especially the services aimed at empowering and integrating visually-impaired people into society, protecting children and raising their awareness against the dangers of drugs, and educating families about this dangerous scourge.

Nae’ma Al Mansouri welcomed the visiting delegation. She praised the humanitarian initiative implemented by the Abu Dhabi Police, and their keenness to provide care and support to people with special needs. She also commended the valuable role played by the ADP to ensure stability, progress and sustainable development for all community members. “The initiative provides blind people with correct information about the drugs scourge, using a writing system that suits them; to shed the light on the negative effects of drug use and abuse, and boost blind people’s culture regarding this danger. As a part of this initiative, brochures will also be distributed to allow blind and visually impaired people invest their time with purposeful activities that raise their awareness about the dangers of drugs,” she explained.

Al Mansouri commended the successful strategic partnership and outstanding relation between the Zayed Higher Organization for Humanitarian Care and the Abu Dhabi Police. “This joint effort is a perfect model that illustrates the outstanding and fruitful cooperation and concerted efforts between governmental institutions, to provide care and assistance to people with special needs and guarantee their right to live in dignity, security and stability,” she said.  She also expressed her admiration for the level of the service provided by the Abu Dhabi police; and for the keenness of the Criminal Investigation Department’s officials to issue these brochures in support of blind and visually-impaired people; and to assist the Zayed Higher Organization in its efforts to integrate visually-impaired people into society. She also commended their dedicated policing efforts to prepare the appropriate environment for blind and visually-impaired people, in a way that reflects the civilized image of the country.

Towards the end of the meeting, Colonel Sultan Suwayeh Al Darmaki presented Ms. Nae’ma Al Mansouri with a commemorative shield from the Anti-Drug Section at Abu Dhabi Police, in recognition of the joint efforts to boost cooperation according to the best international practices.

Colonel Sultan Suwayeh Al Darmaki, Head of the Anti-Drug Section at the Criminal Investigation Department (CID) of Abu Dhabi Police, confirmed that the section has not registered any case of drugs’ use and abuse amongst blind or visually impaired people. He noted that the initiative aims to spread the policing culture, raise awareness about purposeful issues, and strengthen the law respect culture and community values.
